The bacterial cell has protective machinery called restriction- modification system. It has the restriction enzyme and the modification enzyme. The modification enzyme of the restriction- modification system modifies the endogenous host cell DNA by methylation of bases in the recognition site. Thus it will prevent the restrictase from cutting the endogenous DNA.
